Neuronal circuit underlying fine-scale functional columns in the neocortex

One of the major features of the neocortex is columnar clustering of neurons that have similar functions. These columns often have diameters as small as a few somal diameters. For example, in orientation columns, preferred orientations are most similar between neurons that are closer than a few tens of microns in the tangential orientation. In addition, in the mouse brain, we recently found that neuronal activity is similar between subcerebral projection neurons (SCPNs) that are aligned radially within a diameter of ten micrometers.
One possible mechanism of this narrow columnar clustering is common input provided by the same presynaptic neurons. Although a single cortical neuron generally innervates an area much larger than a narrow columnar cluster, it could induce functional clustering if it made strong synapses selectively on neurons in the same cluster.
To test this hypothesis, we conducted simultaneous recording from multiple SCPNs in the mouse visual cortex and examined synaptic inputs provided by the same presynaptic neurons. We found that strong common inputs were largely limited to SCPNs that were aligned radially within a diameter of approximately 15 micrometers. Presynaptic neurons providing strong inputs simultaneously provided weak inputs to distant neurons. These results suggest that selective formation of strong common input underlies functional columnar clustering.
