We previously reported that diabetic polyneuropathy predominantly targets gamma motoneurons of the distal muscle in type I diabetic rats. Alterations in the number and size of motoneurons, however, weren't studied in type II diabetic rats. In this study, we examined the number and cross-sectional area in the medial gastrocnemius (MG) motoneurons of 45-week-old male Otsuka Long-Evans Tokushima Fatty (OLETF) rat and their control strain Long-Evans Tokushima Otsuka (LETO) rat. OLETF rat is an animal model of type 2 diabetes and almost 100‰ of male OLETF rats develop diabetes at 25 weeks of age.
MG motoneurons of OLETF/LETO rats were retrogradely labeled by dextran Texas-Red, and the number and cross-sectional area of cell bodies were examined. Significantly fewer labeled MG motoneurons were found in the OLETF rats as compared with LETO rat. The distribution of mean cross-sectional area of cell bodies of MG motoneurons of both animals was bimodal with optimal threshold between the two cell populations at 600 μm2. The number of smaller MG motoneurons (<600 μm2) was reduced in OLETF rats. As opposed to smaller cells, the number of larger cells wasn't decreased in OLETF rats. Previous studies indicate larger cell bodies were presumed to be alpha-motoneurons and those with smaller cell bodies were presumed to be gamma. We conclude that it is highly possible that diabetic polyneuropathy in type 2 diabetic animals predominantly targets gamma motoneurons.
