Ventral midbrain-evoked rapid monoamine release in the dorsal hippocampal CA2 and surrounding regions

Recent studies demonstrate significant modulation of hippocampal activity and hippocampus-dependent behaviors by monoamines, including dopamine (DA). However, the relationship between functional monoamine release and receptor localization within the hippocampus is not clearly understood, particularly for DA. For example, the hippocampal CA2 is enriched in dopamine D1 receptors relative to surrounding areas, yet there is no evidence detailing functional DA release upon this subregion. To understand the extent and type of functional monoamine input within CA2 and surrounding regions, we recorded rapid monoamine release evoked by stimulation of the dopaminergic ventral tegmental area (VTA) with fast-scan cyclic voltammetry in mice with GFP expression driven by the CA2-specific molecular marker CACNG5. We report here a functional map of VTA-driven hippocampal monoamine release with high spatial precision. In addition, we present pharmacological data that reveals the identity of the evoked monoamine signals (i.e. DA, norepinephrine, serotonin) within different hippocampal recording sites, along with unique effects of modulatory compounds on rapid monoamine release. Our results hold relevance for the precise influence of monoamines within the hippocampal circuit, including CA2, and the modulation of this input by pharmacological interventions in psychiatric conditions.
