Chronic two-photon calcium imaging of layer5 neurons in the mouse primary visual cortex

Mammalian visual system exhibits significant experience-induced plasticity in the early postnatal period. For example, monocular deprivation in early postnatal life induces substantial loss of visual cortical responses to the deprived eye in the primary visual cortex. Previously we revealed that the arrangement of subcerebral projection neurons (SCPNs), a major pyramidal neuron subtype in mouse neocortical layer 5, have a highly organized spatial arrangement. SCPNs align radially to form columnar units, and these units were periodically located. Under specific visual stimulation, neurons in single columnar units exhibited strongly correlated c-Fos expression, suggesting that they have related functions. To reveal the contribution of these columnar units to developmental plasticity in the visual system, we developed a method of chronic two-photon calcium imaging of individual neurons in deep layers. Genetically encoded calcium indicators (GECIs) were expressed in neurons of the mouse visual cortex by using viral vectors. Stable and strong expression of GECIs allowed us to follow individual neurons not only in layer 2/3 but also in layer 5 of the mouse visual cortex. These techniques will enable investigation of the contribution of columnar units to developmental plasticity.
