Association between the GRM7 polymorphism and Beck youth inventories scores in a Chinese cohort

Recent studies have been implicated the polymorphic metabotropic glutamate receptors (mGluRs) to the pathogenesis of schizophrenia. The GRM7, the gene that encodes mGluR7, has been suggested associated with psychiatric disorders. We hypothesized that SNPs in genes involved in the GRM7 are associated with emotional and social impairment in the sample of normal population. Three nonsynonymous SNPs of GRM7 gene were identified on the basis of the information available in the Entrez Gene, HapMap, and Ensembl databases. We performed a pilot study in which we genotyped DNA samples from 20 healthy participants. SNPs with a minor allele frequency greater than 5% in the samples analyzed were identified as the most promising candidates, having representative common variants. The sample includes five hundred and thirty six Taiwanese high school volunteers. Scores from the Beck youth inventories were used to evaluate students' self-concept, anxiety, depression, delinquency, and anger. The result revealed a potential association of a nonsynonymous polymorphism (A/T, rs2229902) in exon 6 with depression. Further analysis detects no significant associations in the other polymorphisms and indices. Future replicated studies with larger sample sizes, and studies that consider different ethnic groups, are encouraged.
