Local ATP generation by dendritic mitochondria and creatine kinases regulates dendrite outgrowth in cerebellar Purkinje cells

Dendrites of cerebellar Purkinje cells develop most complicated branches among all neurons in the brain. Cellular organelles such as Golgi apparatus and mitochondria have recently been shown to play important roles in dendrite morphogenesis. However, the function of local mitochondria in dendrites is obscure. In this study, we analyzed dynamics of mitochondria during dendritic development in cerebellar Purkinje cells. We found that mitochondria were accumulated at the branching points and tips of growing dendrites. The mitochondrial localization in dendrites as well as extension of distal dendrites were disrupted by inducing mitochondrial tethering or by inhibiting TRAK2, while these treatment did not change mitochondrial activity in the soma. Enhanced ATP synthesis by creatine supplementation restored the dendrite arborization in the absence of local mitochondria. Conversely, inhibition of cytosolic creatine kinase (CK) suppressed the extension of dendrites. Growth-retarded dendrites lacking mitochondria showed abnormal aggregation of cofilin. Overexpression of dephosphorylated form of cofilin enhanced cofilin-rod formation and affected dendrite growth. These data suggest that mitochondria in dendrites and creatine-shuttling are important for Purkinje cells to regulate actin dynamics in growing dendrites.
