Distribution of Cortical Projection Neurons in the Rat Subiculum

The topographic distribution of the subicular neurons which give rise to projections to the medial and lateral entorhinal cortices (MEC and LEC), presubiculum (Pre), parasubiculum (Par), and the retrosplenial granular cortex (RSG) was investigated in the rat using retrograde labeling with wheat germ agglutinin-horseradish peroxidase. We found that cells of origin of the projections to above cortical areas were consistently observed throughout the entire septotemporal extent of the subiculum (Sub). While, in the transverse plane, most of the cortical projection cells (i.e., cells projecting to RSG, Pre, Par and MEC) were predominantly distributed in the vertically middle portion of the subicular pyramidal cell layer. The cells giving rise to the projections to LEC were predominantly located in the most proximal (near CA1) and superficial portion in Sub. Only a small number of cells in the deepest (adjacent to the angular bundle) portion of Sub participated to the subiculo-cortical projection. Topographical organization was observed in the projections from Sub to each cortical area along their septotemporal and proximodistal axes. The distribution of cortical projection cells in Sub showed an oblique tri-laminar pattern, which was similar to the previously reported lamination pattern of subcortical projection cells in Sub. These results suggest that such sub-laminar pattern in Sub participate in the function as a watershed of hippocampal output information.
