Top-down information flow is as much a fundamental part of the process of accurate perception as is bottom-up flow. Despite its importance for perception, little is known about the mechanisms of top-down control. We report the identification and characterization of a neural circuit controlled by top-down input in the mouse somatosensory system. The circuit consists of a long-range recurrent horizontal projection between the S1 primary somatosensory cortex and the M2 secondary motor cortex. Physiological recordings revealed that M2 top-down input provides temporally coincident input to the upper and lower layers of the S1, initiating dendritic spikes and selective firing of layer 5 (L5) neurons. Optogenetic inhibition of top-down feedback input in the S1 reduced L5 firing, causing an inaccurate perception of tactile information. These data showed that top-down inputs from M2 to S1 play an important role in accurate sensory perception, which is realized by dendritic and somatic spikes in L5 pyramidal neurons.
