Effects of microinjections of GABAA receptors antagonist into the bed nucleus of the stria terminalis on the intake of conditioned aversive stimulus

We previously revealed that the projections from the basolateral amygdala to anterior part of the bed nucleus of the stria terminalis (BNST) are activated on the retrieval of conditioned taste aversion. Although it has been shown that the BNST receives gustatory inputs, the role of the BNST in CTA has still been unclear. The BNST contains dense GABAergic interneurons, suggesting that GABA in the BNST play a role in the CTA retrieval. Therefore, we examined the effects of the blockade of GABAergic transmission in the BNST by the microinjections of GABAA receptors antagonist into the BNST on the intake of conditioned aversive taste solution during the retrieval of CTA. Initially, rats were implanted with guide cannulae for microinjections into the BNST. After recovery, they were trained to drink water from a syringe during a 20-min session for 5 days. On Day 6, all rats were presented with a 5 mM saccharin solution as a conditioned stimulus (CS), immediately followed by an i.p. injection of 0.15 M lithium chloride (2% B.W.) as an unconditioned stimulus. After this conditioning, on the Test 1 (Day 9), the rats were presented with the CS, 30 min after the microinjection of saline into the BNST. On the Test 2 (Day 11), the rats were divided into two groups so as to match the consumption of the CS on the Test 1. One group was microinjected with saline, the other with bicuculline (100 ng/0.25μl). Both groups were presented with the CS as well as the Test 1. The both groups acquired robust aversion to the CS, because their CS intake on the Test 1 were significantly lower than those on the conditioning. On the Test 2, the saline-injected rats displayed aversion to the CS, but drank more CS solution than the Test 1. In contrast, the bicuculline-injected rats displayed strong aversion to the CS and there were no differences in the CS intake between test days. On the Test 2, the CS intake in the bicuculline-injected rats was significantly lower than those in the saline-injected rats. These results indicate that the blockade of GABAA receptors in the BNST enhance the suppression of the consumption of an aversive CS on the CTA retrieval.
