Stereological estimation of olfactory receptor neurons in rodents

In recent years, the olfactory system has attracted considerable interest because of important discoveries regarding the odorant (olfactory) receptors expressed by the olfactory receptor neurons (ORNs). Olfactory receptors have been extensively studies, and 1035, 1207, 811 and 387 receptors were reported in mice, rats, dogs and humans, respectively. Olfaction is mediated by a huge number of ORNs of the main olfactory epithelium (MOE) in the nasal cavity. Signals of ORNs are then converged to the olfactory bulb and further transmitted to the olfactory cortex in the brain. In this study, the total numbers of ORNs in the unilateral MOE of rodents were estimated by stereological method. Skulls and noses of young adult (8-weeks-old) C57BL/6J mice, and newborn (postnatal day 0) and young adult (8-weeks-old) Wistar rats were decalcified, embedded in paraffin, cut into 10-μm thick sections serially at 100-μm intervals, and processed for immunohistochemistry for the olfactory marker protein (OMP), a specific marker for mature ORNs with fiber connections to the olfactory bulb. The total number of OMP (+) ORNs was estimated by using commercially available stereological software, Stereoinvestigator. The total number of ORNs in the unilateral MOE was approximately 5Χ106 in young adult mice. The corresponding unilateral total neuron numbers were approximately 0.5Χ106 in newborn rats and approximately 20Χ106 in young adult rats. We concluded that rats had 4 times more ORNs than mice at the young adult stage, and that they increased about 40 times between birth and maturity. We will also provide quantitative data on the total number of ORNs in aged rats.
