Various cognitive functions are thought to have close relationships to local field potentials (LFPs) with specific frequencies. However, the frequency property of LFP propagation across cerebral cortex is not clearly understood. To address this issue, we introduced a combination of optogenetics and micro elecrocorticogram (ECoG). We specifically photo-stimulated the deep layer excitatory neurons in primary visual cortex (V1) with various (5-238 Hz) frequencies and compared LFP responses among three sites; the stimulation site (icLFP), cortical surface near to the stimulation site (ECoG Near) and cortical surface far from the stimulation site (ECoG Far). Recombinant adeno-associated virus vector expressing hChR2 (E123T/T159C)-EYFP under control of the CaMKIIα promoter was injected to 8 hemispheres of 5 Long-Evans rats at the depth of 0.5- to 1.5-mm from the cortical surface. After 3-4 weeks, a 32ch ECoG mesh was subdurally attached over the transgene positive site and an optrode was inserted into that site for photostimulation and icLFP recording. Virus injection and electrophysiological recording were conducted under general anaesthesia (injection: Nembutal, recording: urethane). Comparing with icLFP, high frequency responses were significantly reduced in both ECoG Near and Far in the fundamental and harmonic components. Differences between ECoG Near and ECoG Far in these components were less significant. In non-harmonic components, beta- or low-gamma signals were significantly dominant in ECoGs than in icLFP. These results suggested that LFP signals propagated vertically from the deep layer excitatory neurons to the cortical surface with low-pass filtering and modification by the gamma-generating local circuitries. Then they might propagate horizontally in less frequency-dependent manner.
