Parkinson's disease (PD) is a disorder chiefly due to a critical degeneration of dopaminergic neurons in the substantia nigra pars compacta (SNc); The main cause of the PD pathology is considered to be the malfunction whose level ranging single cells to the ensemble across the dorsal striatum (dStr) and others in the cortico-basal ganglia-thalamo-cortical (CBTC) loop. However, the axonal projections from SNc to dStr are too long, complex and experience-dependent to completely regenerate with their massive axon collaterals. As a consequence, several indirect approaches for the alleviation of the PD pathology were developed: dopamine replacement therapies by medications such as L-dopa, or by directly injecting dopaminergic induced pluripotent stem (iPS) cells into the dStr, lesions of regions involved in the CBTC loop such as subthalamic neucleus (STN) or internal globus pallidus (GPi), and deep brain stimulation (DBS) for activating or inactivating the layer V of the motor cortex, STN or GPi by high frequency electrical or optogenetical manupulations. However, such open-loop approaches have not considered the dynamic state of the CBTC loop from single cell to the ensemble levels. A closed-loop approach which adaptively and immediately stimulates the neurons within the CBTC loop in response to the dynamic states monitored by the neuronal activity within it, might markedly improve the amelioration of the PD pathology. Here, we proposed a novel closed-loop DBS system in which simultaneous recordings of multiple single-unit activities and local field potentials (LFPs) from the primary motor (M1) cortex, and optical stimulations to STN or M1 in conjunction with AAV-CaMKII&alpha-ChR2 injected into them were coupled together through a custom-made hard real-time controller. We introduce the core devices realizing the system in detail, and demonstrate results of preliminary experiments in rats of the 6-OHDA-induced hemi-PD model under evaluations using an apomorphine induced rotation test.
