Parkison's disease (PD) shows symptoms of wide variety of psychomotor dysfunctions including bradykinesia, rigidity, and tremor, however, there is no standard diagnostic test for human patients. PD is characterized by degeneration of dopaminergic neurons in substantia nigra pars compacta (SNc), leading to the depletion of dopamine (DA) in the striatum. The depletion of DA in striatum is thought to cause changes of neuronal activities in basal ganglia, which relate to the PD symptoms. However, the relation between the changes of the neuronal activities of each region of basal ganglia and the pathological state of PD is ambiguous. Here we show that the activity of the dorsolateral striatum is correlated to the pathological state of PD. We found in the MPTP intoxicated mice model of PD (PD-mice) that the activity of dorsolateral striatum analyzed by activation-induced manganese (Mn2+)-enhanced magnetic resonance imaging (AIME-MRI) is significantly correlated to tyrosine hydroxylase (TH)-immunoreactivity in striatum, which is known to correlate to the motor performance in PD mice. There is no significant correlation between the activity of ventromedial striatum and the TH-immnunoreactivity. In addition, the activity of the dorsolateral striatum is highly correlated to the fraction of the TH-positive neurons in SNc but not in ventral tegmental area. We also confirmed in the striatal slice preparation of the mice that the amount of accumulation of the Mn2+ is highly significantly correlated to the neuronal activity. Our results demonstrate that the activity of the dorsolateral striatum is correlated to the pathological condition of PD, and that AIME-MRI has the ability to analyze the neuronal activity in vivo non-invasively. We anticipate our results to be a starting point for unraveling the pathophysiology of PD, because AIME-MRI can analyze the activity of the whole brain. Furthermore, our results show that AIME-MRI can be utilized to the researches and the diagnosis for the overall neurological disorders.
