PDGF-dependent regulation of pericyte and neurosecretion in adult mouse neurohypophysis

Hypothalamo-neurohypophysial system releases vasopressin and oxytocin ftom axonal terminals of the neurohypophysis into blood circulation for controlling body fluid homeostasis and lactation. Chronic physiological stimulations are known to cause neurovascular and neuroglial reconstruction in the NH of adult mammalian, no study has been reported for vascular plasticity. In the present study, we aimed to elucidate functional significance of platelet-derived growth factor (PDGF) in pericyte-derived vascular plasticity and neurosecretion in adult mouse. Light and electron microscopic observation revealed that blood-derived tracer molecule, fluorescent isothiocyanate (FITC) and dextran 3k, diffused into neurohypophysial parenchyma at a limited vascular region such as perivascular protrusion, indicating perivascular protrusion is a main route of neuropeptides diffusion. Chronic physiological stimulation with drinking of 2% NaCl increased vascular permeability of FITC and surface area of perivascular protrusion. The area increase of perivascular protrusion possibly resulted from the release of PDGF-B from oxytocin-containing axonal terminals and upregulation of PDGF receptor B on pericyte. Thus, the present study indicates that PDGF-dependent shape conversion of pericytes is responsible for expansion of perivascular surface area and thereby promotes facilitation of neurosecretion.
