Effects of antidepressants on encounter-induced hyperactivity in isolation-reared mice

We recently showed that encounter with an intruder induces hyperactivity in isolation-reared mice and this is associated with a transient activation of prefrontal dopaminergic and serotonergic systems (Neuropsychopharmacology 2013). The novel abnormal behavior hyperactivity was inhibited by drugs which affect serotonin, γ-aminobutyric acid and glutamate systems (Int J Neuropsychopharmacology 2014). To clarify the pharmacological profile of the encounter-induced hyperactivity, we examined the effects of various antidepressants and related drugs on the behavior in isolation-reared mice. An isolation-reared mouse was placed in the large compartment, which was divided by a mesh partition into smaller compartments. An intruder mouse was introduced into the unoccupied small compartment. The behaviors of the resident mouse were videotaped, and analyzed using the ANY-maze video tracking software. Encounter with an intruder increased markedly the locomotion of an isolation-reared mouse and the hyperactivity was repeatedly observed at least for 2 weeks. All antidepressants including the tricyclic antidepressant desipramine, the serotonin reuptake inhibitors fluvoxamine and citalopram and the serotonin/noradrenaline reuptake inhibitors venlafaxine and duloxetine inhibited the hyperactivity in isolation-reared mice without any effect on the locomotion in group-reared mice. In contrast, haloperidol did not inhibit the hyperactivity at the dose which had no effect on the locomotion in group-reared mice. This study shows that encounter-induced hyperactivity in isolation-reared mice is inhibited by various antidepressants. The results suggest that the encounter response of isolation-reared mice is a depressive-like behavior and is a behavioral phenotype sensitive to antidepressants.
