Stem cell technology, using induced pluripotent stem cells (iPSCs) or embryonic stem cells (ESCs), provides us with human spinal motor neurons (MNs), which had previously been inaccessible, to investigate MN diseases including amyotrophic lateral sclerosis. However, limitations including low efficiencies remain in the currently available MN differentiation protocols that hamper the use of human iPSCs/ESCs.
We designed a chemical library screening with the aim of identifying a molecule that promotes differentiation of MNs from iPSCs/ESCs. We validated the screening assay, and set up screening flows using HB9-GFP knock-in human iPSCs/ESCs based on a conventional MN differentiation protocol. This study demonstrates that a phenotypic assay using reporter stem cell lines would be used for a large-scale compound screening for MN differentiation.
