Projection patterns of individual spinocerebellar axons in the mouse

Mossy fibers, the largest class of cerebellar afferents, originate from precerebellar nuclei in the brain stem and spinal cord, and convey sensory or efference-copy information to the cerebellum. Their projection patterns have not been fully clarified because there are many sources of mossy fibers. Among these sources are the external cuneate nucleus, which conveys sensory information from the forelimb, and Clarke's column in the thoracic segments (dorsal spinocerebellar tract, DSCT) and spinal border cells in the lumbosacral segments (ventral spinocerebellar tract, VSCT), which convey information from the hindlimb. We analyzed the projection patterns of individual spinocerebellar axons from these systems in mice, in which axonal reconstruction in the spinal cord is practicable. The spinocerebellar axons were labeled by injecting biotinylated dextran amine into the gray matter of the thoracic and lumbosacral segments of the spinal cord. Entire axons were reconstructed from serial sections of the cerebellum, medulla and spinal cord except in the area near the injection. Axonal terminals were mapped onto the molecular compartments of the cerebellum revealed by aldolase C (Aldoc, zebrin II) immunostaining. The axons from the thoracic projected mainly to Aldoc-negative compartments in the paravermal areas of lobules II-V and lobule VIII, whereas the axons from sacral segments projected mainly to Aldoc-positive and negative compartments in the vermal areas of lobules II-III and VIII. These different projection patterns were partially confirmed by retrograde labeling experiments in which fluorescent tracer was injected into particular aldolase C stripes in the vermis and paravermis of lobule VIII. The results indicate that the functional distinction among different spinocerebellar systems is related to the compartmentalization of the cerebellum.
