Natural variation modifies temperature responses and memory in C. elegans

Temperature is one of the critical information for living and proliferation of organisms on the earth. We are studying about natural variation of genes involved in temperature tolerance of C. elegans. Most of 20 degree-grown wild-type N2 animals isolated from Bristol can not survive at 2 degree, whereas 15 or 17 degree-grown N2 animals can survive at 2 degree. We found that various wild-type strains isolated from variety of area exhibited different cold tolerance phenotypes. CB4854, CB4855, CB4858, RC301 and KR314 can not survive at 2 degree after cultivation at 15 or 17 degree, while N2, CB4853, CB4857 and CB4932 can survive at 2 degree after cultivation at 17 degree. Because CB4854 showed the strongest decrement of cold tolerance, we focused on CB4854 strains. We used deep DNA sequencer and classical genetic mapping to identify the genes involved in the natural variations. Responsible gene polymorphisms for the cold tolerance was mapped on chromosome X, by the classical linkage mapping and three factor crosses. Deep DNA sequencer analysis showed that there are eighty thousand DNA polymorphisms including SNPs between CB4854 and N2. we have mapped responsible polymorphism onto -1.6~ -1.5cM region on chromosome X, where six genes are predicted. We thus tested cold tolerance of the mutants defective in these genes, and we found that vps-52 mutant animal exhibited abnormal cold tolerance that is similar abnormality as CB4854. vps-52 gene encodes Golgi-associated retrograde protein (GARP) that is mainly expressed in neurons. CB4854 has a nucleotide insertion in intron near splicing site. We are now going to do rescue analysis and quantification of expression levels of vps-52 in natural variation strains.
Recently, we are studying temperature memory by utilizing temperature shift experiments in cold tolerance. In N2 wild-type animals, only three hours after temperature shift from 25 to 15°C , cold tolerance was newly established. Our phenotypic analysis demonstrated that natural variation, CB4857, rapidly memorized new temperature. We are planning to identify the responsible genes for the memory variations by using deep DNA sequencer.
