Sex difference of the effect of neonatal social isolation on experience driven synaptic delivery of AMPA receptors in rat neocortex

Although many other reports have discussed sex differences of the response to various stresses until now, however, the molecular mechanisms of these phenomena are still poorly understood. In order to identify these molecular mechanisms, we studied the sex difference of the effect of neonatal social isolation in rat barrel cortex. In this study, pups were isolated from mother and other siblings for 6 hours per day from P7 to P11. We analyzed sensory neuronal network mechanism at P14.
Here we show that neonatal social isolation disrupts experience-driven delivery of GluA1 containing the α-amino-3-hydroxy-5-methylisoxazole-4-propionic (AMPA) receptors to synapse in barrel cortex of males, but this phenomenon is not seen in females rats. Furthermore, we found that injection of glucocorticoid disrupts experience dependent synaptic GluA1 trafficking in the barrel cortex of male but not female rats. Moreover, we showed that disruption of GluA1 trafficking with social isolation was not prevented in masculinized female rats. Our findings suggested that female rats could have molecular machinery which protects them from neonatal social isolation stress induced disruption of synaptic AMPA receptors delivery.
We hope the critical molecular mechanism that is responsible to evoke sex difference will be found, and make it use to improve mental disorders driven from neonatal social isolation stress.
