Hyperglycemia causes the reduction of numbers and sizes of abdominal motoneurons in STZ rats

Diabetic peripheral polyneuropathy (DPN) is a major complication in diabetic patients, but it had been believed that motoneurons are relatively protected from DPN. We recently observed that there is a significant decrease in the absolute number and size of medial gastrocnemius (MG) motoneurons, especially in the smaller motoneurons, in diabetic rats and reported that it is highly possible that DPN predominantly targets smaller motoneurons of the distal muscle. However, also the functional disorder of abdominal (Abd) muscles, postural and breathing disorder, is pointed out in diabetic patients. In this study we examined alterations in the number and size of motor nucleus of Abd muscles using experimental type I diabetic rats.
Type I diabetes was induced in 13-week-old male Wistar rats with streptozotocin (STZ, 50mg/kg in saline; i.p.) administration. The age-matched control animals were injected with saline only. After 2 weeks, each animal was used for labeling of Abd motoneurons. All animals were anesthetized with Halothane (0.5-1%). Abd muscle nerve was sectioned and dipped for 2 hrs into a small polyethylene tube containing 10% dextran-fluorescein dissolved in saline. After 3 weeks survival period, the animals were re-anesthetized and perfused. The spinal cord was sectioned serially, observed with fluorescence microscopy, and the number and size of cell bodies were examined.
The mean number of labeled Abd motoneuorns was significantly less in the 2-week diabetic rats as compared with control animals. The mean soma diameter of Abd motoneurons was also significantly smaller in the 2-week diabetic animals. We conclude that chronic hyperglycemia induce the reduction of number and size of Abd motoneurons in only 2 weeks after STZ injection.
