Area-specific dopamine receptor expression in adult mouse astrocytes

Substantia nigra pars reticulata (SNr) shows very dense dopamine D1 receptor (D1R) immunoreactivity. However, only minimum amount of D1R-positive cell bodies could be detected in this area, leading us to speculate that fine processes might well be responsible for the expression. Indeed, neither parvalbumin-positive nor tyrosine hydroxylase-positive cell body appeared to be colocalized with D1R immunoreactivity in confocal microscopic examinations. To identify the D1R immunoreactivity, we used D1R-promoter-controlled yellow fluorescent protein (D1-YFP)-expressing transgenic mice. Single cells were acutely dissociated from SNr of these mice. Interestingly, strong YFP signal was detected from the processes of putative glial cells in SNr of D1-YFP mice (> 85%). In contrast, neurons in SNr showed no strong YFP signal in their cell bodies, although synaptic bouton-like expression could be detected on them. Double immunostaining using anti-GFAP antibody and anti-D1R antibody further indicated that over 90% SNr astrocytes did express D1Rs in their processes. Similar examination was performed in striatum and visual cortex as well, indicating that approximately 40% of astrocytes and neurons express D1R in striatum, whereas only weak D1R-expression could be detected in a small population of cells in the visual cortex (12.8% in astrocytes, 6% in neurons). Dopamine D2 receptor (D2R)-expression was investigated as well in striatum, SNr and visual cortex. From these, we discuss area-specific dopamine D1 and D2 receptor expressions in astrocytes.
