Transient down-regulation and restoration of levels of glycogen synthase in transected rat facial motoneurons

Transection of adult rat facial nerve leads to a functional down-regulation of motoneurons and glial activation/proliferation. During these events, how energy-producing reactions are regulated in an axotomized facial nucleus is less understood. Here, we investigated the changes of the molecules mediating glycogen synthesis/degradation following the nerve injury. There was no significant difference in levels of glycogen phosphorylase between the contralateral and ipsilateral nucleus during first 2 week after axotomy, whereas the amount of glycogen synthase in leasioned side was significantly decreased during days 7-14 post-injury. A quantitative analysis showed that the glycogen synthase levels in axotomized facial nucleus were reduced to approx. one-half at 14 days after insult. An immunohistochemical result observed that the decreased glycogen synthase is seen in the injured motoneurons. In addition, the glycogen synthase levels in the axotomized facial nucleus had returned to control levels by 5 weeks post-insult, as had the cholinergic markers. The immunohistochemical study also revealed the recovery of glycogen synthase levels at the later stage. Accordingly, our results revealed that the injured facial motoneurons transiently reduced glycogen synthase levels at around 1-2 weeks post-insult, but restored the levels at 4-5 weeks post-insult.
