Shitsukan coding in marmoset visual neural network including posterior superior temporal area

In order to address the cortical mechanism of material perception ("Shitsukan"), we presented a set of stimuli that have identical three-dimensional shapes (bone, torus or amorphous) but have different material appearances (ceramic, glass, fur, leather, metal, stone, wood, or matte) to anesthetized marmoset monkeys (Callithrix jacchus ), and recorded multi-unit activities from its posterior superior temporal sulcus (pSTS) region with 4-shanked silicone probe. In all 4 animals recorded, we found small 400 to 800 μm diameter subregion in the posterior ventral pSTS where most cells showed significant main effect in material appearance (2-way ANOVA, "object shape" x "material appearance", p<0.05), and the best stimulus was gloss material (glass or metal). In 3 animals, we injected retrograde fluorescent tracer to such gloss-selective subregion. One week later, we identified multiple retrogradely labeled spots posterior to the injected site by in vivo fluorescent imaging. Later histological staining from 2 animals confirmed these spots lay in areas MST, MT crescent (MTc) and TEO, but not in MT. In 1 animal, we inserted multiple-electrode array in the pSTS gloss-selective subregion and in one of the posterior labeled region, and successfully conducted simultaneous recording from these connected region while presenting the visual stimuli. 1) In the probe placed in MTc labeled spot, multiunits with main effect to material appearance were found across cortical layers in 2 out of 4 shanks. This effect was vanished only in the deep layer by pixel shuffling of the stimulus images. 2) In TEO, main effect for shape was observed in the superficial to middle layers of all 4 shanks. 3) In the pSTS gloss-selective spot, we found multiunit whose main effect to material appearance and appearance-shape interaction disappeared by pixel shuffling, but the effect to object shape was maintained. These results suggest that information related to material perception is processed within MTc region, and is integrated in pSTS with shape-information processed in TEO.
