The differences of the spatiotemporal patterns between carbachol-induced theta waves and disinhibition-induced epileptic discharges in rat hippocampal slices

Theta and beta rhythms are related to memory and learning process. During the generation of the rhythm, hippocampal cells fire synchronously. Hippocampus has another pathological rhythm in which the cells also fire synchronously. It is an epileptic discharges. In the discharges, amnesia is often induced. In the present study, the spatiotemporal patterns of the current-source density were studied in these rhythms and discharges. The theta rhythms were induced with the application of the cholinergic agent carbachol. The epileptic discharges were induced with the application of GABAA receptor antagonist picrotoxin. The spatiotemporal pattern of potential in the CA3 region of hippocampal slices was recorded using 8×8 multi-electrode array system MED64 (Alpha MED Scientific Inc.). From the results of the potential, the spatiotemporal pattern of current source density (CSD) was calculated using Laplacian filter. As a result, the synchronized synaptic sink and source at the wider area were induced in the epileptic discharges than that in theta rhythms. The magnitude of the current was larger in the discharges than in theta rhythm. It was previously shown that the application of carbachol suppressed the epileptic discharges and the further application finally induced beta rhythm. In the present study, the same results were reproduced, and calculated the CSD spatiotemporal pattern. The results show that the large current induced in the epileptic discharges became smaller with the application of carbachol. The synchronized area and duration of the current in the discharges became smaller and shorter with the application. These results suggest that the area size, the duration, and the magnitude of the synchronized current will distinguish epilepsy and theta rhythm in hippocampus. The difference may be related to the difference of the memory process during the two kinds of the rhythm.
