The role of CAPS2 in adult neurogenesis

Calcium-dependent activator protein for secretion 2 (CAPS2) is known to regulate the secretion of large dense-core vesicle and promote brain-derived neurotrohpic factor (BDNF) in hippocampal neuron. In addition, BDNF has been implicated in regulating adult neurogenesis and their maturation in the subgranular zone (SGZ) of hippocampal dentate gyrus, however, the participation of CAPS2 in adult neurogenesis via BDNF remains unclear. Using CAPS2 knock-out (KO) mice, we investigated the association between CAPS2 and neurogenesis in the SGZ with cell proliferation marker [5-bromo-2'-deoxyuridine (BrdU)] and neuronal maturation markers. In our experiment, adult CAPS2 KO and wild type (WT) mice were injected with BrdU and housed under standard condition or environmental enrichment (EE) condition equipped with running wheels, which is known to promote BDNF secretion and enhance adult neurogenesis in SGZ. BrdU immunopositive cell numbers were compared between CAPS2 KO and WT mice in both control and EE conditions. Furthermore, the maturation of newly generated (BrdU positive) neurons will be analyzed by co-immunostaining of immature neuronal marker, carletinin, and mature neuronal marker, calbindin. In this study, we report the effect of CAPS2 in adult neurogenesis and their maturation.
