Effects of neuroprotective agents on oxidative stress in association with Alzheimer's disease in vitro

Aging is the strongest risk factors of Alzheimer's disease (AD). Oxidative stress is one of the phenomena, which increase in relation with aging. Our group has detected two mediators,IGFBP3(Insulin-like growth factor binding protein)and WISP1(Wnt1 inducible signaling pathway protein 1), released by glial cells when activated by A beta. We investigated the relation between these two mediators and AD. In this study, we examined the effect of these mediators and other neuroprotective agents on cells under oxidative stress and examined A beta production. We used CHO/APPswe-Ncad cells treated with H2O2 as oxidative stress. We added Vitamin E, Nobiletin, Galanthamine hydrobromide, Nicotine, WISP1, IGFBP3, IGF1 with or without H2O2 to the cells and examined cell death using MTT assay and amyloid beta 40 and amyloid beta 42 levels using Human amyloid beta 40 and amyloid beta 42 ELISA kits. Galanthamine hydrobromide, Nicotine, WISP1, IGFBP3 had been shown to be neuroprotective against oxidative stress. Moreover, Galanthamine hydrobromide work on CHO/APPswe-Ncad cells to produce less amyloid beta 40.
